Automicroneedle Therapy System Combined with Topical Retinoid Shows Better Regenerative Effects but Fewer Side Effects on the Skin Compared to Individual Treatments

Abstract
Regenerative therapy has become a new field for dermatologists. Various noninvasive modalities including topical agents have been used to treat depressed scars as well as wrinkles. Several available topical agents for these kinds of skin problems have some limitations of transdermal drug delivery due to their large molecular weight and low liposolubility. Recently introduced microneedle therapy system (MTS) making multiple mechanical holes into the skin improves transdermal delivery. Automicroneedle therapy system (AMTS) is a developed type of MTS and has several advantages compared with MTS. This study was designed to elucidate the cutaneous effects of AMTS with topical retinoid. Twelve hairless mice were divided into 4 treated parts: 1) AMTS with 0.025% tretinoin; 2) 0.025% tretinoin only; 3) AMTS with vehicle; 4) no treatment. Cutaneous regenerative effects and side effects were evaluated by the functional assessment including transepidermal water loss and skin hydration and the histopathology including the epidermal and dermal thickness and the densities of collagen. Western blot and real time RT-PCR were also performed for protein and mRNA of procollagen type I and matrix metalloproteinase-13. ATMS with retinoid revealed better dermal regenerative effects resulted from the proliferation of collagen fibers by increasing tretinoin penetration compared to individual treatments and also reduced the epidermal side effects of topical tretinoin.
